The Neuropathology and Pharmacology of Schizophrenia

چکیده

Despite a well-established role for dopamine (DA) in the neuropathology of schizophrenia, and the evidence of a hyperdopaminergic state in the schizophrenic brain, many questions still remain. Typical agents acting predominantly on DA Dz receptors are only partially effective. lew data now indicate that the interaction between DA and the various DA receptors as well as DA interaction with other transmitter systems, are more critical in deciding the therapeutic success of an antipsychotic than actions on DA alone. These interactions are closely associated with what is being documented regarding the neuro-anatomy, neurobiology and neuropsychology of the disorder.

Effect of Olanzapine and Risperidone on Oxidative Stress in Schizophrenia Patients

Oxidative stress has been suggested to contribute to the pathophysiology of schizophrenia. Increased oxidative stress is the result of either an increased production of free radicals or a depletion of the endogenous antioxidants. However, whether this imbalance of oxidant-antioxidant status in schizophrenia is integral to disease itself or the effect of antipsychotic drugs is not clearly unders...

Animal models of cognitive dysfunction and negative symptoms of schizophrenia: focus on NMDA receptor antagonism.

Cognitive deficits in schizophrenia remain an unmet clinical need. Improved understanding of the neuro- and psychopathology of these deficits depends on the availability of carefully validated animal models which will assist the development of novel therapies. There is much evidence that at least some of the pathology and symptomatology (particularly cognitive and negative symptoms) of schizoph...

Neurochemistry and neuroendocrinology of schizophrenia: a selective review.

Neurochemical investigation has played a major role in the search for the cause of schizophrenia. Initial research strategies involved the direct measurement of neurochemical substances in biological fluids. Subsequently, indirect measures of brain biochemistry including pituitary hormones and responses to pharmacologic probes were examined. Recent advances in in vivo functional neuroimaging, b...

Bridging pharmacology and neurodevelopment in schizophrenia.

At the beginning of the 20th century, when neuropathologists had long searched without success for the neural basis of schizophrenia, this field was called a ‘graveyard for neuropathologists ’. This disappointment opened the door for psychiatrists to focus on psychopathology. The discovery of antipsychotic agents in the 1950s provided a clue to understanding the pathology of this difficult dise...

Comparing the Effects of Adjunct Aspirin and Simvastatin on Psychopathology among Inpatients with Schizophrenia

Objectives: As a severe mental health condition, schizophrenia presents a chronic and complex clinical manifestation and neuropathology. A large body of literature exists on the pharmacological treatment of schizophrenia. However, evidence on some dimensions of such interventions (e.g., eligible candidates, potential predicting factors of the therapeutic outcomes, safe implementation of these i...
